Aushon Protein Array Technology

Aushon Chemiluminescent Protein Arrays are created by spotting up to 12 different capture antibodies in each well of a 96-well plate using piezoelectric printing technology. Samples or calibrated protein standards are added to the wells of the plate, resulting in the capture of appropriate target proteins by the arrayed antibodies. A cocktail of biotinylated antibodies that specifically bind to the captured proteins is then added. Finally, an incubation step with streptavidin-horseradish peroxidase conjugate (SA-HRP), followed by the addition of a luminal-based substrate, results in the generation of chemiluminescence at spots where target proteins were captured.

The concentration of each analyte in the array is quantified by comparing the spot intensities for each unknown sample to the corresponding standard curves calculated by the PROarray Analyst Software. Integrated density values (IDVs) are proportional to the concentrations of bound proteins. Individual analytes are identified by the position of each specific capture antibody within each well. Because multiple assays are conducted within each well, a cooled CCD imaging system and specialized array software are required for Aushon Chemiluminescent Array Analysis. Aushon's new SignaturePLUS system is a complete system designed specifically for this with integrated PROarray Analyst software for unmatched ease-of-use and enhanced sensitivity and dynamic range.
